1) PROTEOLEA, background As prime target, we wanted to develop a product able to ‘recharge’ Proteasomes in our skin cells. Proteasomes, an intracellular complex of proteins that has a tubular shape, are located inside each of our (skin) cells and are able to degrade and recycle damaged proteins, take care of cell- cycle control and regulate Apoptosis (programmed cell death). The complete process is directly linked with the aging process of cells. Proteasomes will lose around 50% activity between the ages of 20 to 60. PROTEOLEA© will be able to counter-act and accelerate proteasomal activity again. Proteasomes, even if extremely tiny, are visible under an electron-microscope. Their existence was already suspected back in 1950 but it is only since 1979 that 3 scientists from Israel (Aaron Ciechanover, PhD), Hungary (Avram Hershko, PhD) and the USA ( Irwin Rose, PhD) succeeded to shed light on the mystery of the Proteasomes . After 25 years of in-depth research, the 3 scientists were then awarded in 2004 with a Nobel prize in chemistry for their amazing discoveries. They had unraveled that Proteasomes play a key role in regulated protein degradation which is a step-wise process: damaged proteins are labeled, recruited to the Proteasome, absorbed and then recycled into peptides and amino acids, which can then serve as building blocks for new proteins. Back in 2000, Prof. Chondrogiannii from Greece could also make a link between active Proteasomes and aging. He had discovered that healthy centenarians showed Proteasome activity which was closer to that of young people than to elderly ones. We then had decided to link these pharmaceutical findings into cosmetic application. 2) PROTEOLEA, concept history Until recently, action of Proteasomes has long been neglected by the cosmetic industry. Whilst we had started with an idea in 2006, major development of PROTEOLEA© was done in 2010/11. Thanks to the experience we already had with its sister-product ILLUMISCIN© (Proteasome action on Lipofuscin pigments, patent of RAHN AG), we could develop and launch PROTEOLEA© in just 2 years. PROTEOLEA© offers a holistic approach to anti-aging and is a clever complex of the following components: o Taylor-made olive leaf extract Everything started with educated guess. Whilst different oils and fatty acids were reported to offer some positive action in activating Proteasomes, we have discovered after various in-vitro tests that olive leaf extract offered the best activation of Proteasomes, thanks to Oleuropein. A major hurdle was however how to implement an Oleuropein-rich olive leaf extract into cosmetics, given its notorious poor solubility and dark color. We could then develop a special extract after lengthy testing that offered the properties we were looking for. Main action of our special olive leaf extract will be to activate the Proteasomes as such. o Jujube extract To support the action of olive leaf extract, we were looking for a first line of defense, namely reduce damage on cellular proteins as prophylaxis. We are firmly convinced that Jujube seed extract, rich in Jujubosides, offer a very powerful anti-oxidative wall on the skin. Additionally, Jujube fruit is also known in Asia as to be about 100 times richer in vitamin C than conventional sources like citric fruits. Jujube, as Asian plant, offers a perfect fit to European olive leaf extract. o Levan To complete the Jujube- (first line of defense) and olive leaf extract (2nd line of defense), we wanted to raise a third line of defense that should compose a film on the skin surface and also offer a good skin feel. Different screenings of long-chain Polysaccharides showed that Levan was offering what we were looking for. Levan is by the way already well known by the cosmetic industry for its nice skin feel. Levan as an inert material is however not key in reducing wrinkles since it has no biological function, contrary to olive leaf extract that shows measurable, long term sustainable effect. Still and as a positive side effect, Levan will offer the ability to reduce wrinkles in the short term. - PROTEOLEA© today The synergistic approach of PROTEOLEA© (PROTEasomes + OLEA (Olea euopae = olive) will offer you an uncommon solution in reducing wrinkles for an age group going from 30 to 50. A very large study on 80 ladies has shown that PROTEOLEA© was able to reduce age by o 6 years in just 4 weeks, with just 3% concentration, with detailed back-up data Besides protein recycling, Proteasome activation is thought to improve the so-called cell cycle control which means that: o Population of younger skin cell will increase and will take less time to shed off o Cells will have less junk (damaged proteins) inside and will be more functional than older cells o Cell death (Apoptosis) will be delayed As slight increase of skin renewal by 10-20% will be beneficial, according to practical experience. This may also be true for stem cells which might enjoy extension of life span. Pools of stem cells will therefore be less depleted as a result.
