Varenicline tartrate is a partial nicotinic agonist that acts on α4β2 nicotinic receptors. It alleviates withdrawal discomfort but also diminishes rewarding effects of cigarettes smoked if patients lapse when using the drug.Standard varenicline tartrate dosing (a gradual increase to 2 mg/d) was formulated to avoid adverse effects (primarily nausea) in sensitive patients. The latest findings suggest that the limits to treatment response to varenicline may be due to factors other than insufficient dosage.
