Summary from today- we used confal microscopy to show that when compared with a control, molecule x bound onto m2 macrophages. The experiment was set at 4 degrees celsius to negate the physiological ability of macrophages to engulf our molecule x. This step was important as the confal microscopy results are needed before we can use flow cytometry to determine whether the results from flow cytometry where due to binding, or internalisation of molecule x by the macrophages. Flow cytometry was used to quanitfy the binding, and when compared with controls, confirmed that molecule x specifucally bound to m2 macrophages but also bound to other cells. We concluded that molecule x therefore mediates immune responses through binding to m2 macrophages with high specificity, activating or changing their function leading to a th2 anti inflammatory response.
